Krangean (LITSEA Cubeba (LOUR.) The press.) Is a plant from familial Lauraceae and spread abundantly throughout tropical and subtropical Asia, including Indonesia. Krangean has many benefits that can be used in traditional medicine, such as toothache and antitoxin bite insects. Previously, Xantin Oxidase (XO) inhibition test from Krangean tree skin extract was evaluated. This study aims to determine the inhibitory activity of XO methanol extract of Krangean stems and isolate marker compounds. Research starts with characterization and phytochemical screening of raw drugs. Krangean extract is obtained in methanol with a reflux, filtered, and crowned method. It was monitored with thin layer chromatography (KLT). Extract was also tested for the XO inhibition activity. Fractionation is done with vacuum liquid chromatography and classic column chromatography. Subfraction is purified with Preparative KLT, and the purity is examined with a single Development and two-dimensional development. Isolates monitored with KLT with potassium hydroxide and dragendorff spray reagent. Isolates show blue fluorescence under ultraviolet light (UV) 365 Nm and orange color under visible light. Furthermore, it is characterized by the visible UV ray spectrophotometer. It has a maximum wavelength of 228, 281, and 301 nm. Based on these characterizations, the isolate is thought to be a Kumarin compound. At 100 g / mL, extract inhibits XO activity of 22.52 � 1.88%. The maximum resilience concentration (IC50) of the extract is 229.27 g / ml.
